Indomethacin and Juglone also modulate the expression of several key genes and proteins involved in the regulation of apoptosis, cell cycle progression, oxidative stress, and inflammation. Indomethacin induces the expression of caspases and Bax, and inhibits the expression of Bcl-2, leading to the induction of apoptosis in cancer cells. Juglone induces the expression of Nrf2, a transcription factor that regulates the expression of antioxidant and detoxification genes, leading to the reduction of oxidative stress in cancer cells. Preclinical and clinical studies have shown promising results for the use of Indomethacin and Juglone in the prevention and treatment of colon cancer. In preclinical studies, both compounds have been shown to inhibit the growth of colon cancer cells in vitro and in vivo, and to suppress the development of colon tumors in animal models. Clinical trials have also shown promising results for the use of Indomethacin and Juglone in the prevention and treatment of colon cancer, with minimal side effects.
