This book presents an efficient in vitro propagation protocol for Gloriosa superba Linn., an endangered medicinal plant, and Plumbago indica L., a rare medicinal plant. Optimized culture conditions successfully promoted shoot induction and multiplication in both species. For G. superba, MS medium supplemented with 4.0 mg/L BAP and 1.0 mg/L IAA produced shoots in 92% of cultures, with an average of four shoots per culture. For P. indica, MS medium containing 0.5 mg/L BAP resulted in shoot production in 95% of cultures, with six shoots per culture. The addition of casein hydrolysate, activated charcoal, and coconut water further enhanced shoot multiplication, producing up to 12 and 14 shoots per culture, respectively. Species-specific auxin treatments also promoted successful rooting of in vitro raised shoots. The protocol offers a practical and promising approach for the rapid propagation, sustainable utilization, and conservation of these valuable medicinal plants.
